TL1A-Responsive Luciferase Reporter Cell Line
TL1A (Tumor Necrosis Factor-like Ligand 1A) protein is expressed in various immune and non-immune cells, including T cells, monocytes, macrophages, dendritic cells, and endothelial cells. TL1A has two main functions: first, it can induce apoptosis and inhibit angiogenesis,which may limit tumor growth and making it a valuable cancer target. Secondly, TL1A can stimulate inflammation and fibrosis by activating certain immune cells and fibroblasts. TL1A binds to two receptors: death receptor 3 (DR3), which provides stimulatory signals for downstream signaling pathways; and decoy receptor 3 (DcR3), which inhibits TL1A activity.
TL1A is involved in the development and pathogenesis of many inflammatory and autoimmune diseases, such as Crohn’s disease, rheumatoid arthritis, psoriasis, primary biliary cirrhosis, systemic lupus erythematosus and ankylosing spondylitis. TL1A enhances the production of proinflammatory cytokines, such as IFN-γ, TNF-α and IL-17, by stimulating TH1 and TH17 pathways. TL1A can also activate fibroblasts and induce fibrosis in affected tissues.
The market for therapeutics for autoimmune diseases is huge, and many companies are actively researching TL1A as a target, especially for inflammatory bowel disease. For example, Merck recently purchased Prometheus Bioscience for nearly $11 billion dollars, primarily for its anti-TL1A antibody, PRA023, to treat ulcerative colitis and Crohn’s disease. Meanwhile Pfizer has teamed with Roivant Sciences to make its own anti-TL1A drug, RVT3101. Both products are in early-stage clinical trials.
BPS Bioscience has developed a TL1A-Responsive Luciferase reporter cell line, #78811, that stably expresses DR3 and an NF-κB promoter that drives luciferase expression upon activation by TL1A. This cell line provides a simple and sensitive method to measure TL1A activity and its effects on DR3/NF-κB signaling. The cell line can be used to screen for TL1A agonists or antagonists, or to study the role of TL1A in inflammatory and autoimmune diseases.
