FLAER Alexa Fluor® 488 Proaerolysin
Cedarlane offers FLAER Alexa Fluor® 488 proaerolysin, Used as a flow cytometry reagent for PNH (Paroxysmal Nocturnal Hemoglobinuria) diagnosis and research.FLAER is an Alexa Fluor®488 labelled inactive variant of aerolysin, a unique protein that binds tightly and specifically to mammalian GPI anchors. FLAER does not bind to PNH cells. It is this reliable absence of FLAER binding to GPI anchor deficient cells that allows this reagent to efficiently detect leukocyte PNH clones. Before FLAER, detection of PNH clones by flow cytometry relied on fluorescent labeled antibodies to GPI-linked proteins such as CD59(MIRL) and DAF(CD55).

Highly Specific GPI Anchor Binding - Binds tightly and directly to glycosylphosphatidylinositol (GPI) anchors on mammalian cell surfaces.Superior Sensitivity vs. Traditional Methods - More sensitive than CD59 and CD55 antibodies; even very small PNH clone populations can be detected that would be missed by conventional antibody-based methods.Reduced Risk of False Negatives - Traditional antibodies screen for loss of a specific protein (e.g., CD59 or CD55), leaving room for false negatives. FLAER targets the GPI anchor itself - if the anchor is absent, cells are FLAER-negative.Validated for Multi-Parameter Flow Cytometry - Used alongside antibodies (CD45, CD33, CD24, CD15, CD14) to detect PNH clones in monocyte and granulocyte lineages per clinical guidelines.Available in Two Formats - Offered in both liquid and lyophilized formats for lab flexibility.
